Its hard to tell with some experience you can tell. The way to see is if they look like they are going to explode and you can usually see the vein in their back pulsating.
It just depends on how long you want to wait to get your colony going. That way would get you going in prob like 6 months at a minimum but prob longer. Id start with like 80 females and 20 males would be better but i went with the cheaper wait longer method and its just starting to get going.

You should be misting I would say 3 times daily and your not misting him your misting his cage. Mak sure to cover all the leaves well and dont spray them if they dont like it some do some dont it jsut depends on the chameleon and possibly how fine the mist is
some chameleons are just that way and never change but the best you can do is slowly try and hand feed and if he gets good at that then try to lure him onto your arm. Just dont force anything or youll stress him out too much
