Could a male and female Panther chameleon live comfortably together in a 16"x6'x6' enclosurer?

No, honestly two panthers would need to live in an enclosure at least the size of a good sized room in order to get along and be able to get away from each other, it would also have to be fairly well planted.

Even with an enclosure that size you would really have to know what the signs of stress for the male and female are, and be able to separate them before any real health issues arise.

Males have been known to attack females in enclosures, when it's not mating time. Domestic violence.
